Tramplers can trample anything that is smaller than them. Cavs are size 3, eles size 6. Ergo, elephants can trample cavs.

Note that damage is partially affected by size difference, so horses can't be trampled quite as effectively. Chariots are size 4, so they are near useless against cavalry as tramplers. And of course, Mammoths are a nightmare for Hoburg, once they reach(until then, crossbows.)

Damage is only affected by size difference in that AP is reduced more for trampling larger opponents, so the total damage doled out by a single trample sequence will be less if the opponents are large.
Damage on a single successfully trampled unit is only determined by the size of the trampler, not the size difference.

Yes, but he AP value is very important, and what it actually works out to is more damage on smaller creatures are you rush them, with relative size pretty critical.

Don't forget the def check to reduce the damage to 1 (capped)
In theory units with lighter armour should be a sensible counter against elephants. Particularly light cav with their mounted boost to def, for example. But it isn't really the case, in practice.

Also keep in mind that mistform units don't appear to take the 1 damage if they dodge the trample, but merely get moved around a bit. I think that they still get the -2 def penalty per trample attempt for the rest of that round (i.e. -2 def every time they are attacked in 1 round) but damn is it nice to not take ANY damage from trampling until they are successful. Just make sure to stack up that def on the mistform units as I've seen one just disappear when his def finally runs out. I forget if he took 1 damage and lost mistform when he got trampled successfully the first time, but I imagine that's how it'd work (and then he gets squashed on the next successful trample). This was 1 vanjarl vs. 12+ elephants so there were many, many trample attempts.

I generally see a lot less trample damage to high def units... they just get pushed around and take a 1 HP damage. I've used thugs with high def to rout elephants/etc.
